Sclerotinia in Brassica napus is caused by the fungual Sclerotinia sclerotiorum, which is one of the most three serious diseases in our country.lt is significant to study on the mechanism of diseases resistance against S. sclerotiorum. To value the resistance of the two varieties line, S.sclerotiorum was inoculated to the plants grown in field. After inoculation of the fungus, mycelium growth conditions were observed. We set 12h、24h that after inoculation as the key experiment point.By compare the protein expression profile in a resistance line with that in its relatively susceptible one during the interaction of B.rassica napus with S.sclerotiorum,6512 different peptides,i.e.998 proteins were founded differentially expressed by using iTRAQ quantitative technology, among those different proteins, a number of important proteins were related to disease resistance. In order to identify the putative funtion of those differentially expressed proteins, we performed BlastX alignments against NCBI non-redundant databases Swiss-Prot, COG,and K.EGG,G0,KEGG databases. All those proteins are related to different metabolic functions respectively. We verified the most important proteins. With RACE technique or PCR cloning, total sequence of BnBRI1、BnRLK5 and BnPEPR2 were obtained. After analyzing tissiue specificity of BnBRII,it was found that expressions in roots and flowers was highest, whereas they were lowest in seeds. We built phylogenetic tree of BnRLK5 and BnPEPR2.PEPR2 can recognize peps to trigger immune responses. BAK-a co-receptor, which regulates multiple signa ling pathways, may play an important role in PTI.
