Arbuscular mycorrhizal fungi(AMF) were widely distributed and could form mutualism with terrestrial plants. Through two-way transfer of resources, the partners overcome all kinds of biotic(pathogens and herbivores) and abiotic(drought) stresses.The symbiosis between AMF and plants were a fine molecular regulation process,fromed pre-symbiotic signaling dialogue to plant stress defense and cell morphological remodeling in symbiosis stage, every step involved many biological molecular.Different from traditional mode plants, Lotus japonicus and Medicago sativa which were herbs, in this study, Amorpha fruticosa as woody leguminosae plant who may harbor slightly different symbiosis mechanism was selected as the host plant, at transcriptional level to investigate the molecular mechanisms underlay the symbiosis between AMF and Amorpha fruticosa.RNAseq method was employed to screen the differential expressed genes in Amorpha fruticosa roots induced by AMF during the colonization process. 2460 differentially expressed genes were obtained by transcriptome sequencing, with 881 genes up-regulated, 1579 genes down-regulated, In addition to the unknown function genes, only be predicted genes, And when the differential expressed genes satisfy the following conditions: Fold change>1,FDR(q-value)<0.001,RPKM>20. We obtained285 differentially expressed genes, of which 82 genes up-regulated, 203 genes down-regulated,According to GO classified information, Blast annotation and related literatures, the 82 up-regulated genes were assigned into seven categories including stressand defense, metabolism, energy, protein synthesis, signaling, transcription related,protein folding and degradation related. In according to screening criteria was Fold change >2, we obtained 50 significantly down-regulated differential expressed genes.Furthermore, for q RT-PCR validation, and 4 of these genes were successfully confirmed with the result of RNAseq, show that sequencing result was believable.The colonization of AMF on Amorpha fruticosa was a system activity involving many biological processes, all aspects of the body function unit work together to realize the final goal of mycorrhiza formation between AMF and Amorpha fruticosa,facilitating the exchange of nutrition.
