Part I Effects of β -Casomorphin on gastrin release of gastric antrum 1. Superfttsion system is established and then employed in the experiment as an approach to investigate the role of β -Casomorphin, in gastrin release of 10 days' piglet and its mechanism, to explore the dynamic effect of β-Casomorphin, on the gastrin release of newborn piglets of different days. Superfusion system is established and employed in the experiment. Gastric antrum of 10 day& piglet can be stimulated by β -Casomorphin, (2* i0 10 5molIL) to secrete gastrin and this effect cannot be reversed by naloxon. β - Casomorphin3 (2*1 0ol/L) shows no effect on the secretion of gastrin. β - endorphin(2* 10~ molIL) can stimulate the secretion of gastrin through non-opiate mechanism. Casein (l4gIL) hydrolysate also possesses this ability while casein fails to evince such effect. Reduced glutathione (GSH, 2*10-mol/L) can also stimulate gastrin release. Medium with or without 10% newborn calf serum shows no apparent difference on gastrin release. Naloxon alone also has no effect on gastrin release. The effect of stimulating gastrin release by stimulants such as β - CM, manifests desensitization. β -CM, (I 05mo1/L) exerts strongest effect of stimulating gastrin release on 1 day piglet, and the piglet of 28 days shows delayed appearance of desensitization. 2. Effects of β -Casomorphin on gastrin release of mucosa cell of gastric antrum in primary culture. Medium with β -CM, (2*10 1 OSmol/L) was collected after having treated antral cell for 3 or 12 hours respectively. β -CM, can stimulate antral cell of rnucosa to secrete gastrin via non-opiate mechanism. 3. Effects of infusing β -Casomorphin, into newborn rats and mature mice and of injecting it into the belly cavity of newborn rats on the gastrin level in serum. Infusing β -CM, (10~5molIL, 0.5mLI animal) can enhance gastrin release in 10 daysSD rats fasting for 8 hours while injecting β -CM, (10~molIL, 0.2mLI animal) induces opposite effect. Infusing β -CM, (1 O5moIIL, 0.5mLI animal) can also promote gastrin release in mature mice fasting for 12 hours in the same time β - CM3 fails to produce any effect. Reduced glutathione, however, evinces even more potent effect than β -CM,. Infusing β -CM, has no effect on gastrin release in mice being fed with soya-bean cake. Part II Effects of ~ -Casomorphin on the proliferation of anterior Pituitary cell in primary culture β -CM, (2* 10ol/L) can prompt the proliferation of anterior Pituitary cell of 10 days piglet via MTT method. β -CM, also has the ability to stimulate the proliferation of anterior Pituitary cell of newborn piglet of different (1, 5, 28 ) days via 3H-TdR method, and the effect is most pronounced in 5 dayspiglet. Part 111 Effects of ~ -Casomorphin on the transformation of lymphocyte in 10 dayspiglet and the natural proliferation of lymphocyte in newborn piglets of different days β -CM, (2*10-7, 2* 105mo1/L), Casein (14g/L) hydrolysate and β -endorphin (2*1 08mo1/L) can prompt the transformation of lymphocyte in 10 dayspiglet, and naloxon cannot reverse the effect. β -CM, can stimulate the natural proliferation of lymphocyte in 1 day and 5 dayspiglets. The overall results elucidate that β -Casomorphin, can stimulate gastrin release of piglets , prompt proliferation of anterior Pituitary cell, stimulate natural proliteration and transformation of lymphocyte. β -Casomorphin,, as the major bio-active peptide produc...
