In order to reveal the genetic diversity of Zea mays sinesis in the Southwest of China and accumulate some experimental data for the study on the origin and evolution of Zea mays sinesis, nine Zea mays sinesis populations and five Zea mays indurata populations were studied. Three means including morphology observation in field, isozyme, and SSR makers were applied in the study. The results as following: I .There were distinct differences between Zea mays sinesis and Zea mays indurata. Although the genetic diverge also exits within Zea mays sinesis populations, the trend is the difference among Zea mays sinesis is smaller than that among Zea mays indurata populations. The materials from GuangXi and YuNan displayed strong photoperiod sensitivity. 2.Six different peroxidase zymograms were obsered among 14 populations. The difference between Zea mays indurata revealed by isozyme makers is not very big. All materials?maps has t the fifth band. This band can be regarded as the signal band of germplasms origined in China. 3.SSR research indicated that good polymorphism of SSR map which showed the high differential ability of the primers. Especially, primer bnlg 1292 can be the special SSR primer used for distinguishing Zea mays sinesis and Zea may's indurata. The cluster analysis based on binary data the true genetic difference existence between Zea mays sinesis and Zea may's indurata from the view of molecular. The difference not only exsits in wax gene and its correlation sites, but also scatters in the maize genome. The relative size of genetic difference can be indicated by simple matching coeffeicent. 4. Because of the significant difference between Zea may's sinesis and Zea may's indurate revealed by morphology observation, rozyme, and SSR makers, the hypothesis that Zea may's sinesis origined from a single gene of Zea may's indurate is proved not correct. The original and evolutional route may be known by some new methods such as rDNA's ITS analysis. From above all, it can be summarized that a significant genetic difference exists between Zea may's sinesis and Zea may's indurata and the genetic diversity of the both is very abound. The results from this study are useful for maize theory research and breeding work.
