Question about taxonomic status of Pinus den4flora var. ussuriensis Pinus dens flora var. sylvestrzformis is being discussed by many researcherS for a long time, but there is no a specific conclusion now. In this article, PCR- RFLP and DNA sequence techniques are used to study rbcL gene of cpDNA of Pinzss sylvestris var. mongolica Pinus dens4lora var. ussuriensis Pinus dens flora var. sylvestr~formis Pinus tabzdaeformis var. mukdensis Pinus thunbergii in northeast regin of China. taxonomic status of Pinus deizsjflora var. ussuriensis Pinus dens flora var. sylvestrjformis and systematic evolutions of five two-needle pines in northest are revaluated. (X) According to study of two extraction methods (CTAB and SDS), qualification and quantity of DNA from CTAB method are better than SDS method among two-needle pines. ?Product lengths of first and second PCR are same among five two-needle pines. It suggests length of rbcL gene is very conservative , there are no gene insertion lost and rearrangement. ?According to analysis of four restriction endonuclease maps, it suggests P. dens flora var.. sylvestrjformis P. dens flora var. ussuriensis P. sylvestris var. mongolica are same in maps, P. Labzdaefiz,rtnit varmi&lensis P. thwibergll
