| Molecular Cloning and the Recombinant Fowlpox Virus expression Vector Construction of the SI Gene of Infectious Bronchitis VirusIsolated in XinjiangWang wei (Veterinary Microbiology and Immunology)Supervised by Pro.Shan wenlu, Mr.Kong xiangang and Mr. Liu shengwangAbstractTo study the molecular epidemiological background and genetic background of infectious bronchitis virus isolated in Xinjiang and to develop effective new type vaccine, from Xinjiang Urumqi we isolated a field strain,(IBV-XJ3),from flocks which broke out lB. By electron microscope observation~. animal regression experiment and haemagglutination inhibing nature study etc. We confirmed the XJ3 isolate was IBV. The immune gene Si of IBV-XJ3 was amplified by RT-PCR,the product(1658bp) was cloned into pUCI9 plasmid and confirmed with blue/white dot screenings restriction enzyme analysis and PCR. After nucleotide sequencing ,amino acid sequence was deduced and compared with 8 foreign reference strains and 5 Chinese isolates selected from GENEBANK by software GENERUNNER~ CLUSTAL X and TREE VIEW, the results were:IBV-XJ3 had less than 80% similarity with 8 foreign reference strains, 87% similarity with JX199/Oi,96% similarity with vaccine strain QX. Less than 80% similarity with other Chinese isolates. Proved IBV-XJ3 was varriant different from foreign reference strains. Then we constructed a recombinant fowlpox expression vector containing the Si gene of IBV-XJ3.The research has a great significance in the exploration of molecular epidemiology of IBV and developing genetic engineering vaccine. |
