The main factor limiting the efficiency of the utilization of urea by ruminants is the rate at which urea is hydrolyzed in rumen. Urea is converted to ammonia and carbon dioxide and the resulting ammonia is frequently produced more rapidly than the microorganisms can utilize it for amino acid and protein synthesis. One solution the problem of the rapid rate of ammonia release is relayed by adding urease inhibitor to ration.Five trials were conducted:In trial 1, In vitro the simulating experiment was conducted with strained rumen liquor. theureaseinhibitorconcentratingon0~ 6X1LY3~ 6X10~ 6Xl0~. 6X10~. 6X107~ 6X810 M was incubated with strained rumen liquor and urea buffer for 1 h at 39 C ,and thenammonia concentration was tested.Trial 2 was conducted on effect of UJ on ammonia releasing velocity & pH value. A mono-factor design was adopted to investigate the optimal addiction of UI in diet in vivo. Twelve lambs equipped permanent fistula were allotted randomly to 4 groups of 3 animals (replicates) which were treated with administration of 0, 20, 40, 8Omg/kg,diets. The experimental period consisted of a 7-day preliminary period followed by a 2-day collection period. In trial 3 Degradation rate of DM, NDF, ADF was determined by nylon bag technique and the same design was adopted. Also did the same diet and experimental period. In trial 4 DMD, CPM,NDF and ADF of diet were determined by total feces urinary collection method in a 3 X 4 mono-factor design. Experimental period consisted of a 10-day adjustment period and a 5-day collection period. In trial 5, Twenty weanling crossbreed lambs (Texel X local breed) were allotted to five group of 3 animal (replicates) with a balanced average body weight in all the groups. Each group was feeded urea-rich diet (1%, 1.5%, 2%urea of complete ration) +UI, basic diet +UI and basic diet without UI.The experimental results were shown that:I It was optimal in vitro for Ul to inhibit the activity of urease when the concentration was 6 Xl 0~mo1/l. More or less was not effective mostly.2 The experimental result in vivo showed that the abrupt release of ammonia did not occur by adding UI 20,40,80mg/kg to treatment diets. The urease was decreased by 45.5%, 62.2%, 44.0% respectively (p
