Screening of the Somaclonal Variation in Watermelon(Citrullus lanatus L.) and Its AFLP AnalysisAbstractIt is hypothesized thdt Fusarium oxysporum is a possible cause of Watermelon wilt. Fusarium oxysporum mycelia proliferated on a large scale in Watermelon transfusion tissue lead to block in the vascular system, thus resulting in illegitimate hydrate metabolism.And Fusarium oxysporum mycelia produce a toxin named fusaric acid (FA) in host plants, which can injure plant cell structure, lead to disorder in metabolism and cell desiccation, therefore resulting in plant wilt.In this paper, FA-resistant plants were obtained using a watermelon tissue culture system. Watermelon variety inxia?was used as material and putative somaclone mutants were obtained by first being induced by FA and then screened in vitro.The putative mutant were analyzed using AFLP silver staining. And it was confirmed that when using Primer M2E1, the obtained PCR products composed of a specific DNA band of FA-resistant Watermelon plants.However, whether the wilt-resistant capability of FA-resistant watermelon somaclone mutants is consistent with their FA-resistant ability has not been determined yet. Therefore further field wilt-resistance experiment is still required.
