Three sugarcane varieties were used for the genetic transformation by using Agrobacterium tumefaciens LBA4404 containing LEAFY gene and selectable marker gene of NPT II and GUS. Calli derived from the leaf of sugarcane were infected and cocultured with Agrobacterium tumefaciens at logarithmic phase. The highest transformation frequency was 9.09% according to the estimation of Kan resistant calli produced . The putative transformed plant were confirmed by Gus assay, PCR analysis and PCR-Southem blotting. This transformation procedure of sugarcane will provide an efficient mode for the transformation of monocots.The LEAFY gene was also introduced into cells of sugarcane calli using PDS-1000 He particle bombardment. The callus was cultured on the media with Kan(300mg/L). 7 clones with regenerated capacity were screened out on the media.The result of obtaining the transgenetic sugarcane plants provided the scientific proof for studying the expression of LEAFY gene in variety plants and made good fundament for inducing sugarcane blossom by gene engineering.
