Random amplified polymorphic DNA (RAPD) were used to analyze the genetic distance and genetic diversity of six local pig populations in China, including: Qianbei Black (Guizhou province), Licha Black (Shandong province), Yimeng Black (Shandong provinee), Laiwu (Shandong province), Bamei (Shanxi province) and Bama X i ang p i g (Guangx i province). Of thirty-six random primers used to amp I ify, twenty-six random primers generated polymorphic fragment, each with 6.23 fragments. The polymorphism frequence of each primer ranged from 20 to 100%. A total number of 162 reproducible RAPD fragments was generated from the 26 primers, of which 105 were poIymorphic, and the total polymorphism freqence was 64.81%. The freqence of polymorphic markers was 55.24%, 53.33%, 49.52%, 47.62%, 46. 67% and 45. 71% in Qianbei Black, Licha Black, Bama X iang, Yimeng Black, Bame i and La i wu p i g, respectively. The genetic distance index between populations calculated by Nei ranged from 0. 1220 to 0. 3099. The closest genetic distance was observed among Licha Black, Yimeng Black and Laiwu pig(0. 1220 ~ 0.1786). The farthest genetic d i stance was found between Bama X i ang and Bame i, Li cha BI ack pig, La i wu or Y i meng BI ack p i g (0. 2571 ~0. 3099). Q i anbe i BI ack p i g had c I oser genet i c distance to Licha Black, Bama Xiang and Laiwu pig(0. 1813M). 1879), but had farther genet i c d i stance to Y i meng BI ack, Bame i p i g (0. 2335?. 2457); The genetic distance index between Bamei and Licha black, Yimeng black or Laiwu pig ranged from 0.1910 to 0.2209. The phylogenetic tree was constructed by Neighbor joining (NJ) method. NJ tree showed that Yimeng Black pig and Laiwu pig was grouped to one cluster, which was firstly grouped with Licha Black pig, then all that was grouped with Bamei pigto construct one unit; Qianbei Black pig and Bama Xiang pig was grouped to another unit; Final Iy, these two units were grouped to one cluster. It was not observed that two individuals have entirely identical pattern of RPAD bands whi le genetic diversity were analyzed on Qianbei Black pig and Licha Black pig . The genetic similarity index was 0.8354+0.0129 and 0. 8348+0. 0063 in Qianbei Black pig and Licha Black pig respectively. No RAPD analysis had been reported on these six local pig populations previously and the results from the present study warrants further research on the classification, conservation and exploitation of these pig populations.
