Mannitol-1-phosphate dehydrogenase (mtlD) gene from E.Coli, was successfully transferred into upland rice by biolistic method. The condition of callus culture of upland rice was inquired.The experiment results indicated that: 1. The proper hormone concentration for mature embryos is 2.5mg/L2.4-D or 2mg/L2.4-D+0.5mg/LABA. 2. To upland rice, the proper cyt for differentiate is 6-BA, and the suitable rate of NAA/6-BA is 0.2-0.3. 3. The frequent of microprojectile bombardent can influent the selecting rate and regeneration ratio of resistance callus. Two times of bombardment had better effect than one or three or more. 4. The amount of inherited can influent the induction rate. 14 days is the better one. 5. The molecular test indicated that the positive ratio of regenerated plant is 70% or so. 6. 80 percent of the positive plants can tolerance 0.5% NaCl.
