It is important to analyse New centuary, Hongfeng and other excellent and new apricot cultivars using RAPD for propagating and breeding furthermore. In this research, studies have been taken on 20 apricot cultivars. We have established and optimized the DNA extract and RAPD amplification conditions. The fingerprints and specific bands of Hongfeng, New centuary and other studied cultivars have been found out. The pedigree and relationship among them have been studied. The main results are as follows:1. Apricot DNA extraction and RAPD amplification conditions have been established and odptimized.Simplified way using CTAB is rapid and high efficient to extract DNA that can be used in RAPD amplification from apricot leaves.Spring leaves are the best materials to extract DNA. More DNA can be extracted using SDS way than DNA extracted using CTAB way, but in the way of SDS, more impurities can be contained in extracted DNA which caused RAPD to fail. In the course of extraction the phenol substances in the leaves of apricot is apt to be oxidized, and so P- HSCH2CH2OH must be added timely to prevent oxidize.The amplification procedure that is appropriate to apricot is as follows:94,2 min;94 ,30sec; 36 ,40 sec; 72 ,lmin;45 cycles;72 5min. The optimized amplification conditions are as follows: The amount of Tap enzyme needed in amplification is 1.0-1.5U, the concentration of Mg2+is 1.5-2.5mM. Under this condition, the stable results can be achieved.2. 18 Primers have been selected from 100S serial primers that can amplify stablely. The amplification results using these primers of 20 cultivars shows polymorphism. 215 bands have been amplified altogether, among which 108 bands show polymorphism, amounting to 50.2 percent.3. DNA fingerprints of Hongfeng, New centuary and other excellent and new cultivars have been established firstly. Awailing of different panels of primers, according to PNA fingerprints and specific bands 20 cultivars can be distinguished.4. 20 cultivars have been analysed by statistics software according to similar coefficient. When the LD is 0.4, the studied cultivars can be classified into the following seven categories:I :Hongfeng,New centuary,HongheBao,Erhaacao V :katyII :Badanshuixing VI:Mano III:Dezhouguoxing.Yuxing,Luotuohuang. VII: Zuoshixing.IV:ItalianNO.lIt can been seen that:The relationship of Hongfeng, New centuary , Excellentl, No.A, NO.B, NO.C, and their parents Honghebao and Erhuacao are very close. The hereditary differences among them are little. This shows the pedigree of them.Great hereditary differences exit in Italian NO.l and katy which come from European cultivars which cause them to be classified into different category.MaihuangXing, Chuanzhihong, Yangjiyuan and Liguangxing are ancient cultivars, The genes of them are conservative, and they belong to the same category.
