These essential component, hemagglutination, toxicity, immunogenicity of Mycoplasma hyopneumoniae strain XY-1 membrane protein were studied. The range of membrane protein MW is from 14KD to 100KD by means of sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and molecular weights of the protective antigens were 36KD and 64KD by SDS-PAGE. Hemagglutination and hemagglutination inhibition of pig red blood cell with Mycoplasma hyopneumoniae strain XY-1 membrane protein at 4# verified hemagglutination of membrane protein, hemagglutination and hemagglutination inhibition titres were 1:27 and l:28.Mycoplasma hyopneumoniae membrane protein was inoculated on chick embryo monolayer cells, using three methods to treat membrane protein. Experimental results verified that membrane protein could generate cytopathic effect, and the minimum membrane protein density generating cytopathy was Img/ml. T lymphocyte transformation test justified that Mycoplasma hyopneumoniae was not only nonspecific mitogen but also specific mitogen. 6 weaning pigs were immunized with membrane protein, and their antibodies were 1:23.50(immunized after 7 days), 1:26.33(immunized after 14 days) and 1:27.83(immunized after 21 days). Humoral and cellular immunity tests testified immunogenicity of Mycoplasma hyopneumoniae strain XY-1 membrane protein was good.
