For studying the distribution and changes of nerve growth factor (NGF) in diencephalons of chicken and exploring the relation between the NGF and reproduction, 5 female healthy chicken of the groups of 120,240,360,and 540 days old were examined by immunohistochemisty SP method, the results of expression of NGF in chicken laying period are as follows:The NGF immunohistochemisty (NGF-IR) positive neurons distribute extensively, they are found in diencephalons everywhere. There are three types of NGF-IR positive neurons, intensely stained cells, mediumly and lightly stained cells. The NGF-IR positive fibers and terminals with varying density and reactive intensity existe in some nuclei. We presume that produce and secretion of NGF had some mode, including target tissue, cell self and side cell secretion and conveyance.In the diencephalons of chicken of 120 day, NGF-IR positive neurons are found only in Nucleus rotundus and paraventricular nucleus,etc. When they're 136 day old , a lot of positive neurons and fibres were found in all position of diencephalons, including Nucleus habenularis medialis, Decussatio supraoptica dorsalis, Nucleus geniculatus lateralis, Nucleus rotundus, Nucleus principalis precommissuralis,Nucleus paraventricularis magnocellularis,Nucleus infundibuli hypothalami,Nucleus periventricularis hypothalami, Nucleus dorsointermedius posterior thalami, Regio lateralis hypothalami,etc, and kept the tread from 136-day old to 240-day. After 240 days, the number of positive neurons and fibres reduced gradually .On the 360th day, some nuclei disappear, when on the 540th days, PVN and ROT etc, are found to have a few of lightly stained positive neurons. The results suggest that NGF has some relation with reproduction.The positive cell density showed a trend of "low-high-low". Before chicken laying eggs, the density of positive cells is low, when startling laying eggs, the intensity become high, on the 240th days, it reach the highest point. In the following days, it decreases. When chicken stop laying eggs ,the cell density go to the lowest point. We could learn from the results that the number of positive neurons may depend on reproductive state.In the egg-laying period of chicken, intensity of positive neurons show a trend of "weak-strong-weak". At the beginning of laying cycle, it was weak, as time go on , intensity became strong, at 136 day, the highest point at 240 day .After the day ,it reduces gradually. On the 540th days old group, intensity of positive neurons fell to the lowest level. We conclude that the secretion of NGF has a strong power on reproduction condition.The NGF-IR neurons and fibers were found in ependyma area containing CSF and show that NGF regulate hypothalams reproduce endocrine by nerve-body fluid. |
