Analyses on genetic diversity and wing polymophism on Rhopalosiphum padi were conducted in this study.1 Genetic diversity was analyzed on 7 Rhopalosiphum padi populations collected from wheat from Beijing, Nanjing, Shanghai, Zhengzhou, Shenyang, Fuyang and Changsha by RAPD-PCR. There are 15 primers among 57 arbitrary primers amplified clearly and specifically polymophic bands. By cluster analysis the 7 populations were grouped into 3 distinct branches. There were significant similarities among collections from Beijing, Shenyang, Shanghai, Zhengzhou and Nanjing, so they clustered on a branch. However, a large distance did exist between the Anhui population and Changsha population, so they clustered on a single branch respectively. The relationship between the result and long-range migration of Rhopalosiphum padi in our country was discussed in the study.2 Researches on wing polymophism of aphids have been widely conducted since 1960's. These researches have concentrated on the induction of environmental factors and the regulation of hormone on aphids' wing development, the results of which indicated that photoperiod, temperature, crowding, nutrition and juvenile hormone as factors could affect on the occurrence of alate/apterous polymophism in aphids. In this study, for the first time, DDRT-PCR technique was applied in analyzing the difference on gene expression between alate and apter in Rhopalosiphum padi. Combining four anchored primers and 20 arbitrary primers, one positive fragment was determined named DD-1. The result of sequencing and aligning with related genes in GENBANK showed that DD-1 was 293bp and homologous to at the least 30 transposes from many sources on sequences of amino acid. The potential function of DD-1 was deduced and discussd.
