The Wheat (jing-411) was cultured with different iron sources (EDTA-Fe, FeCl3, Ferric citrate) treatment and Fe-deficient treatment. First they were used in these experiments to study the differences of the leaf photosynthetic pigment content, the root activity, the root and leaf ATPase activity, nitric acid deoxidizase activity, redoxase activity, EDTA-Fe3+ase activity, four hormones(IAA, CK, ABA, C2H4), the iron absorption kinetics, peroxidase activity and isozyme electrophoresis; second, they were used in these experiments to study the differences of the cell ultrastructure by electron microscope and the root tip surface by Scanning electron scope; third, the gene of avr9 is used in these experiments, including the Southern blotting Northern blotting and RT-PCR. Fourth, the root tip apoprtosis of wheat in different treatments was studied by DNA Laddering electrophoresis and observed by fluoroscope. The results indicate that there are differences among morphologicaK physiological and cytological moleculor aspects in four different treatments. Though the wheat leaf photosynthetic pigment content which be treated by EDTA-Fe ( II ) is higher than others besides the root activity, the root and leaf ATPase activity, nitric acid deoxidizase activity, redoxase activity, EDTA-Fe3+ase activity, the iron absorption kinetics, peroxidase activity, the wheat ATPase activity was treated by Fe-deficient is higher than other treatments. Morepver,the result of isozyme electrophoresis indicated the differences in the root isozyme expression.In addition, the results of electron microscope and the root tip surface observed by Scanning-electron scope also indicated that there are speical differences in different treatments. Finally, the gene(avr9) exists in the genome of wheat, and begins to express in the seventh day and to express highly in the ninth day .
