| The actin cytosckeleton in laticifer wounds and actin in exuded latex of mature rubber tree, Hevea brasiliensis, were studied by using TRITC-phalloidin labeled fluorescent optical microscopy and Western-blottings. The results showed that the actin cytoskeleton in laticiferous cells could be preserved in the materials fixed in 4% paraformaldehyde and 0. 5%glutaradehyde in phosphate buffer(0.1 mol/1 PBS,pH7. 2) and embeded in paraffin in 55癈.It was observed that a mass of TRITC- fluorescent substance appeared in laticifer wounds when latex flow stopped. The fluorescent substance might be the actin cytoskeleton in the protein network of the laticifer wounds. By using Western-blotting , actin was found mainly in C-serum(cytoplasm of laticiferous cells)of the latex collected by exploitation. During a exploitation, latex exuded at the beginning had higher concentration of actin and the actin concentration became lower in latex exuded later.Stimulated exploitation was made with macrolides and KI, which are depolymerizing compounds of actin cytoskeleton. The results showed that application of 1% KI or statured macrolides solution in water led to increase of latex yields, but higher concentration of KI caused serious harm to the bark of rubber tree. |
