The experiments of serial nuclear transfer and cloned embryo transfer for inter-species reconstructed embryo were conducted ,based on Boer goat-rabbit inter-species cloned embryo was obtained by use of fresh goat ear fibroblast cells at 100% cofluency as donor and rabbbit oocyte as recipient. The aim provided the foundation in theory and practice for the improvement of animal clone technology by researching the difference of early development and the possibility to develop in vivo on both inter-species original and serial reconstructed embryos.Study on serial nuclear transfer in goat-rabbit inter-species cloned embryo , reconstructed eggs 858,reconstructed embryos 440,the rate of fusion 51.3%,by culture in vitro ,155 morulas and 55 blastocysts were obtained ,the rate of blastocyst covered 10.2%.Whereafter,73 I serial and 20 II serial reconstructed eggs were gained by use of the blastomere of original cloned morula and I serial cloned morula as donor and rabbit oocyte as recipient. Selected 145 original cloned eggs,then cultured them with 73 I serial and 20 II serial reconstructed eggs together, got reconstructed embryos were 90,58and 20 respectively,namely the rate of fusion was respective 62.1%,79.5% and 70%.The result showed there was no significant difference in the rate of fusion among 3 groups(p>0.05).As for the rate of early development on embryo and the rate of blastocyst ,three were ,72.2%, 75.9% &28.9% ;and10%, 13.8% &0 respectively. There was also no significant difference in the rate of early development on between original and I serial embryo ,However, I serial group was a little higher than original group on the rate of early embryo development .On the contrary , II serial group was the least , reached no morula and blastocyst . On the speed of development, I serial embryos advancerd was faster compared with original group,there was above lor 2 days gap from the fusion to the blastocyst stage . developing to term , the formerwent on 4 or 5 days ,but the latter spent 5or 6 days .There was a key difference in the first 24h after culturing in vitro , I serial reconstructed embryos developed 8-16 cells stage ,but original embryos only reached 4~8 cells stage ,and then the two recovered the same lever again.Study on cloned embryos transfer , 2 groups were divided: original and serial transfer group, 43 recipients, 90 cloned embryos , no cloned offspring was gained. In original transfer group ,the number of transferred cloned embryos was 78 . six recipient goats stop estrus for last 30 days or so after being transferred embryos , even one reached 60 days and represented a serial pregnant reaction .After all ,nothing was found in the uterus , but the uterus in transferred part was thicker than the other by celioscope . In serial transfer group, serial cloned embryos was 12, two recipients didnt estrus until 30 days later after being transferred, however, progestin in blood serum didn t keep a high lever all this time through measure .The result indicated that inter-species original cloned embryos could implant , but serial cloned embryo didn t do it after being transferred in vivo.
