There are scare resistant resources that can be used directly, so it is difficult to gain disease resistant breed by traditional breeding method. Genetic engineering has provided a new way to solve this problem. Using function genes in the process of the signal transduction to induce natural defense system in plant can improve the disease resistance to high level, which have the advantage of broad spectrum disease resistance. According to gene homologue, we have cloned NPR1 gene from arabidopsis using PCR method. Then we construct plant expression vector (pNPR1) with CaMV35s promoter. We introduced NPR1 gene into tobacco. And after PCR,Southern hybridization analysing and disease resistant detect, we gain some highly disease resistant tobacco plants. The result suggest that NPR1 gene have the function of broad spectrum disease resistance. Using the vector pNPR1, On this basis, using the pollen tube pathway, after PCR and disease resistance detect, we gain some verticillium resistant cotton lines. At the same time, we introduced NPR1 gene into colored cotton 9804 with Agrobacterium mediated transformation method and gain plenty of Kna resistance cotton callus and improve the transform system of colored cotton from the age of explants and density of Kna. In order to prolong time of resistance and not to affect the growth and development of crops, we have cloned pathogen induced Pr1-a gene promoter and construct vector pIPR. After transforming cotton with the pollen tube pathway, we have got some Kna resistant cotton plants.
