Protein palys an important role in the animal nutrition. In essence , the nutrient functions of protein is AAs,especially the essential amino acids. Protein needs to be digested free AAs or small peptides and than be absorpted in animal's intestine, so the investigation of the absorption of AAs is always the hotspot in nutrimental fields. The process of the protein digested and the producets uptaked into epethc cells is a persistent dynamic course. So study the course is a very valuable thing to nutrient research .Purpose: At present, intestinal absorption of AAs had various of reports to mouse, rat, rabbit, domestic animal, birds and mammalian small intestine,but few research in marine lives.In order to know the orderliness of AAs absorption in intestine to marine lives,we investigated the kinetics of the 10 Essential Amion Acids(EAA) of fish that its be absorpted and the kinetics of L-leucine and L-Isoleucine that be inhibited by L-lysine, L-valcine, L-Methionine, L-Phenylalanine and L-Isoleucine( or L-leucine) in intestine of Ictalurus Punctatus ,that is our first important porpuse . at the same time ,we hoped that we could supply some useful base datas and found a brief and effective method .Contents: The study focus on the absorption and transport about 10 free amion acids and the effects of L-leucine and L-Isoleucine inhibited by 5 other structure style's AAs and the method of those researches. Three experiments were carried out in isolated gut from Ictalurus Punctatus. The object of the present work was to confirm five questions: (l)we discussed the researchful methods about the amino acids uptaked by intestine; (2)The difference of 10 EAA absorpted and transported were discussed about its in isolated gut from Ictalurus Punctatus;(3)The influencing factors need to be found out to 10 EAA in absorption and transportation and determine kinetic constants of 10 KAA absorpted in isolated gut from Ictalurus Punctatus; (4)Discussing kinetic character of the two mixed AAs be absorbed and transported in isolated gut from Ictalurus Punctatus;(5) The difference of the single AA and two mix AAs in absorption and transportion was compared.Experiment One: The experimental methods of determine the absorption and transportion of AA in isolated gut. The method simulates the physiological condition in the vivo, according to the change of AA solutions inside and outside the gut through the gut, we determined the A A absorpted and transported across the gut wall indirectly. We carried through the experments to Ictalurus Punctatus in this method, The result indicats:(1) the technique has preferable delicacy to measured the AA.,the least quantity is could be precised about 0.5ppm. Based this,we can find out the variational rules accurately.(2) when this experimental system was used ,we had better control the concentrationsfrom 1 to 10mmol/L and the irrigateing time in 60 minutes.Experiment two: The kinetics absorbed and transported research for 10 essential amino acids in isolated gut from Ictalurus punctatus. This experiment was carried out in perfusing single AA solution in isolated gut and mensurating the quantity through ninhydrin test, according to the first experimental system, we determined the kinetic process of 10 EAA absorbed and transported,compared the difference of 10 EAA absorpted and transported one another, The results indicated:(1) From 0 to 21 minutes,the absorbed AAs is increased acording to linear correlation along with the time; From 0 to 60 minutes,there are some difference between the AA transported, in the first 20 minutes, Arg, Lys and Val is more quickly than the latter 40 minutes, it shows that the slope of the 20 minutes' curve more sharply than the latter,But the variety of Leu, Ile, Thr, Trp and His was reversed ,Phe and Met have the same shape curves in the 60 minutes that the change follows the linear correlation.(2)There was a remarkable difference in AA absorbed and transported along with the AA concentration changing.
