Content: A solid static Ivermectin liposome (IVM Liposome) was prepared firstly using Lecithin, Cholesterol, Vitamin E, TW80, and IVM. In order to reduce the cost of liposome, Lecithin soled on the marked was purified by alumina column. The liposome was produced by Ultrasonic- Membrane Method. Its final size was determined by use of dynamic light scattering. The number of bilayers of the vesicles was tested by electron microscopy; negative staining was performed, using 2% Ammonium Molybdate. The mean size of liposomes is 96nm, and the shape of the liposome is round bilayer vesicle. IVM was extract for lipid bilayer by methyl alcohol and determined by HPLC-UVD. The calibration curves for IVM was liner in a range of l-100ug /mL, r=0.9998, n=8. Recoveries ranged from 98.2 to 100.6% (n=5) for ivermectin with 0.5-2.25mg/mL fortified samples. The coefficients of variation were less than 13.9%. the production preserved more than one year at 4 C.Two group of ewe (n=5) were given the formulation at 0.2ng/kg by intravenous administration and subcutaneous administration respectively. The time for collection blood or milk samples was defined base on preliminary test. The recovered plasma samples were extracted with acetonitrile and cleaned up by basic alumina column and determined by HPLC-FLD. And the drug was extracted form milk sample by a series of liquid-liquid extraction steps derivatized with trifluoroacetic anhydride and determined by liquid chromatography with fluorescence detection. The calibration curves for ivermectin in milk and plasma were liner in a range of 0-800ng/mL. Recoveries ranged form 87.0 -102 (n=5) for ivermectin in plasma with 0-800 ng/mL fortified samples and 82-103% (n=5) in milk with the same concentration fortified samples. The limit of detection was 0.04ng./mL for plasma samples and 0.3ng/mL for milk samples. The plasma and milk drug concentrations determined were calculated by software 3p97. In the group administrated the formulation by intravenous route, milk/plasma ration is 1.82, and for the group administrated by subcutaneous route, the ration is 1.2.
