The principle and method of RAPD is applied in this study, twenty six random primers were selected to analysis the excellent strains west bee genome DNA which come from inside and outside of nation at the same time, and according to produced polymorphic DNA of map ,some respect process were selected to calculate and analysis genotype similarity(FK genetic distance(D), genetic diversity(Ho)> genetic differentiation(Gst) et al.The aim is to compare the difference between native province west bee breeds with foreign excellent strains at molecular level, in order to advance the breeding and popularizing of native province. And then improving the fine trait and the quality and quantitative of honey bee production in native province, and providing some theory foundation at molecular level.Through the research, it is show that ,the following RAPD primers have a obviously special band in difference west bee strains, such as sbsJ-04, sbsJ-15, sbs 1224-102, sbs!224-104, et al in the twenty six primers. These primers can be used to differentiate the difference strains of west bee; there have some genetic diversity, but as a whole, the genetic diversity is not high among difference strains of west bee. the genetic diversity level or genetic variation is caused by the gene flow and other factors among the breeds, but the factors within the breeds is small relatively; the diversity among populations is not high in the strain have been researched, average genetic differentiation show that there has 47.51% diversity among populations, all under 50%, and shows that the genetic differentiation is low from the same origin relatively; the average genetic distance is 0.2093 in the eighteen breedsof west bee through the analysis of genetic distance and UPGMA dendrogram also shows that genetic distance is smaller from the same origin, the average genetic distance is bigger from two different strains, the dendrogram can be divided to two branch, one represent the fourteen populations of Apis mellifera linnaeus of royal jelly products % Apis mellifera ligustica Spin, Apis mellifera carnica pollmann and hybridizebreeds. another branch including four population of Apis mellifera caucasica Gorb. at the same time ,it is show that there have a bigger genetic diversity from different strains of west bee.The analysis of native Apis mellifera ligustica Spin shows that there have some degree genetic differentiation phenomena among populations, the genetic diversity comes from the gene flow among the breeds, yet there have a bigger genetic differentiation among native different regions, these results show that there have many types genes that can affect different production performances in native province extensively. There have many high yield and fine quality genes, and they have a wider space and showing occasion, it just need us to make well the breeding and selective mating works.The study about genetic characteristic in different strains of west bee from molecular level, it provides some valuable theory for the honeybee breeding of native province and how to protect and explore the native excellent strains.
