Taurine is an abundant sulfur-containing amino acid found in all tissue types involved in a variety of functions. Taurine was found the highest concentrations in the germen and germ cell, which indicated that taurine is contacted with reproduction. In present study, three experimentations were designed to explore the effects of taurine on the reproduction and its mechanism in male rat.In the chapter one, the author designed and synthesized one pair of primers with the Oligo6.0 system based on the known liver CSD cDNA of rat. A 468 bp cDNA fragment of CSD gene was amplified by RT-PCR from the total RNA of rat testis. The author first confirmed the possibility of CSD gene expression in rat testis, which indicated that taurine can be synthesized in the testis of rat. By the sequence analysis with BLAST 2 SEQUENCE system, the partial nucleotide sequence of CSD cDNA in rat testis was proved to be 99.8% homology with that of liver which was already known, and the putative encoded-amino acid sequence in rat testis was exactly the same as that in rat liver(100%).In the chapter two, the effects of taurine on the secretion of FSH, LH, T and ? were investigated by adding different dose of taurine and B -Alanine to drinking water in Wistar male rat. The results indicated that taurine can stimulate the secretion of FSH, LH, T and E2obviously(P<0.05) , but has no effects on the secretion of E2(P>0.05).In the chapter three, the effects of taurine on testosterone secretion were studied by culturing rat Leydig cells in vitro. The results indicated that: taurine can stimulate testosterone secretion of cultured rat Leydig cells in vitrosignificantly(P<0.01) . The effects of taurine on the testosternone secretion stimulated by HCG showed a biphasic pattern. In other words, testosternone secretion increased significantly(P<0.05) in low concentration of taurine, but decreased significantly(P<0.05) in high concentration of taurine.The results of present study indicated that taurine has many affinities with reproduction of male. And the results will be helpful to farther study the effects of taurine on the male reproduction and its mechanism, and will be useful for its application of taurine in farms and human life.
