The article studied the purification of Rubisco, preparation of the antibody of Rubisco and Photosynthetic rate and the activities and contents of Rubisco were traced during diurnal change and effect alternation of light and dark treatments. The results were abstracted as followings.1. Rubisco was highly purified. Rubisco was obtained from wheat leaves by ammonium sulfate precipitation and ion exchange (DEAD-TOYOPEARL-650M) liquid chromatography within a day. Purified Rubisco, which showed one band by PAGE and two bands at 55,000 and 14,800 by SDS-PAGE, was qualified as antigen.2. The antibody of Rubisco was obtained by immunizing rabbits though in-skin, multi-spot, interval injection.3. Rubisco was determined though simple agar diffusion which was efficient and easy to operate and.4. During diurnal changes of photosynthesis, the initial activities of Rubisco a showed a linearly significant correlation with photosynthetic rate. The diurnal changes of photosynthetic rate and initial activities of Rubisco appeared as a curve with a peak at noon in the mild and bright day and two peaks and a midday depression in the hot day with dry wind. It is a linearly significant correlation between photosynthetic rate and initial Rubisco activity.5. Under alternation treatments of light and darkness, the results showed that light facilitated to synthesize Rubisco ; the activity of Rubisco was activated by light and decreased very fast under darkness.
