Pigeon Paramyxovirus Type I (PPMV-I) strain Sza was propagated in the allantoic cavity of 11-day old embryonated SPF chicken eggs. The allantoic fluid of PPMV-I Sza was purified and concentrated by centrifugation with 10-60% continuous sucrose gradient and was used as antigen to immunize 8-week old Balb/c mice. By using the hybridoma technique, 3 McAbs were established, which were named McAb/C3-f4 McAb/C5-G10 McAb/C6-H8 respectively. Among them, McAb/C3-f4 and McAb/C5-G10 can react with NDV Genotype VI strain Sza and Genotype VII strain YG97. While McAb/C6-H8 react only with NDV Genotype VI strain Sza. The HI titers of the 3 McAbs are low in test, while they are different in VN experiments. The indirect ELISA developed by McAb/C6-H8 was used to screen the allantoic fluid of NDV, and the results showed that McAb/C6/H8 react only with NDV Genotype VI strains, while not with the other strains.
