In the traits of meat, the lean meat percentage and the quality of the muscle are two important target traits in swine breeding programs. Great progress has been made in these two traits, especially in the selection of the lean meat percentage through routine breeding programs, but genetic progress is very slow. With the rapid development of molecular biological techniques, we hope to find some new strategies that can control growth and the potential quality of muscle by the studies of the control mechanisms of the skeletal muscle myogenesis, and then we can use these strategies to alter the muscle growth of the meat-animals. These provide us important theoretical significance and potential utilized value in increasing lean meat percentage and the quality of the meat. This experiment have cloned the MRFs families梩he main myogenic regulatory genes and OB gene?the main gene in affecting fatty deposition in animal, these Complementary deoxyribonucleic acid fragments were amplified by RT-PCR with primers designed according to Gene Bank. The recombinant plasmids pMD18-T-Myog pMD18-T-MyoD pMD18-T-Myf-5 and pMD18-T-OB were constructed respectively. The linked products were transformed to competent cells of E.coli host strain DH5a. Individual clone was cultured and the recombinant plasmids were prepared as template for automatic sequencing. DNAsis and PROsis sequence software analyzed the nucleotide sequences. We had found that the homology of the sequences of Myogenin is 100% among Junmu No.K Taihu pig and the reported gene in Gene Bank, is 100% between Junmu No.l and Taihu pig, but the homology of the sequences of MyoD is 99.5% between Junmu No.l and the reported gene in Gene Bank, which has three mutations in Junmu No. 1 and only one mutation leads to different amino acid, In addition, we had found two different coding sequences on Myf-5, one has 100% homology with the reported gene in Gene Bank and terminate coding at No.600 base in the No.l exon, the other mutated at No.600 base in the No.l exon and can coding the Myf-5 gene completely. With the homology compared, the homology is 85% between Homo sapiens and swine, and is 86% between Murine and swine. In the end, we found two different mutants Genotype on OB gene. One has deleted three bases at No. 143 to 145 bases in the coding sequence of OB gene, which leads to the deletion of No.49 Gin in the amino acid sequence of OB gene. The other hasmutated at No.364 in the coding sequence of OB gene, it causes a mutation atNo. 122 Ala in the amino acid of OB gene.
