Newcastle disease is a highly contagious and economically most important disease of poultry.The disease also inflicts many other avian species.the disease varies in degree of severity ranging from mild,inapparent infecion to severe disease .Based on the severity of the disease in chickens,the causative agent of the disease,Newcastle disease virus(NDV),is divided into three pathotypes : (1) the lentogenic strains,which cause mild inapparent infection ; (2) the mesogenic strains,which cause mild respiratory symptoms with low mortality ; (3) the velogenic strain ,which are highly virulent and cause high mortality. Vaccination has been the widely used method of prevention of this disease .Genetic vaccine is quite different in structure from traditional ones.the experiments show that delivery of genetic material into an animal's cells can trigger some synthesiztion of the encoden proteins as well as of antibodies targeted against those proteins.In order to study immunization of genetic vaccine by inoculation, two kinds of DNA vaccine of NDV are constructed and analysed their sequense, which will express F protein in eukaryotic cell(Hela cell) in vitro with the method of transfection by cationic liposomeThe results testify that two sorts of plasmids DNA successfully expressed the F protein of NDV in Hela cell,but the expressed content of pc4.0F is higher than ones of pc3.1F.We immunize the BALB/c mice with pc4.0F and pc3.1F by paunch and muscle administration and immunize each with different content of 10 g, 50 g 100 g . The antibodies in serum against Newscastle disease virus F gene protein are tested by ELISA. The Result showes that two kinds of DNA vaccines can induce specific antibody response to encoded proteins in 15 weeks by im and ip,but antibodies response is little different.In addition,We successfully construct prokaryotic expression plasmid pTAT-D26F in order to study antigen or protein fused with TAT transduction domain across the intestinal mucosa and transdermal delivery.
