Strain ZM-95,classified into Bovine Viral Diarrhea Virus(BVDV) ,is isolated from the tissue sample of the suspect Classical Swine Fever case. In this study, 11 cDNA overlapping fragments, which covers the complete genome of ZM-95, are amplified by RT-PCR and nest-PCR. Afterwards each fragment is cloned and sequenced. The result shows that the complete genomic sequence is comprised of 12,227 nucleotides. which contains one large open reading frame encoding a polyprotein of 3901 amino acid residues. The 5'untranslated region(UTR) of ZM-95 consists of 337 nucleotides and is at least 35 nucleotides shorter than the 5'UTR of other pestiviruses analyzed so far. The 3'UTR of ZM-95 is 187 nucleotides long and contains a eight-nucleotide repeat sequence, TGTATATA. This repeat sequence is found in 3'UTR of some BVDV isolates, such as NADL, Osloss, and SD-1, but absent from the 3'UTR of CSFV and BDV. No insertion in the NS2-3 region of the ZM-95 was observed.Sequence comparison with other pestiviruses, such as BVDV type 1 strain (NADL and Osloss), BVDV type 2 strain 890 and other genotype strains, revealed that the overall nucleotide identities of ZM-95 were 60.6%, 60.0%, 413% and 37.9%, respectively. However, the deduced amino acid sequence identities of ZM-95, with the genotype strains mentioned above, were 85.0%, 82.4%,68 6%and 64.4%, respectively.The viral glycoprotein E2 of ZM-95 showed lower identities of both nucleotide and deduced amino acid sequence. And the nonstructural protein NS2-3 region is more conserved. All of the comparisons revealed that the ZM-95 has the highest identity with BVDV-I and the genomic structure was the most analogical. This suggested that the strain ZM-95 was classified into BVDV-1.This is the first report about the complete sequence of BVDV isolating from a swine.
