Arabidopsis and Brassica napus both belong to Cruciferae. It had been studied that the whole genomic homology between them is high. According to comparative mapping, the genetic resource of Arabidopsis could be first used to accelerate the improvement of Brassica napus.Recently, several genetic linkage maps from different segregation population of Brassica napus are constructed with probes of DNA clones from Brassica and EST clones from Arabidopsis, and a set of genes that related to economic traits were mapped onto genetic linkage groups. The next step for fine mapping and gene cloning are much difficult, because of the complicated genome of Brassica napus. In this study, we constructed an Arabidopsis expression map with 118 Arabidopsis EST clones, according to the bioinformation and DNA resource in Arabidopsis database. A set of DNA clones from Brassica, which related to important economical traits, was mapped onto the Arabidopsis expression map. According to the results, we confirmed the target fragment in Arabidopsis genome to study Born efficient gene, with the bioinformation in the target section in Arabidopsis for further mapping in Brassica, we found that the molecular markers in the target fragments are colinearity in two genomes. The clones, relevant to the 24 RFLP markers that related to heterosis, were sequenced, and the homologous genes were found. These genes were ascertained to mostly be the signal transduction factors and proteinase in metabolism. At last, we had a discussion on the comparative mapping and the analyzing of heterosis performance on molecular level.
