Study On The Immunogenicity Of 54KD Outer Membrane Protein Of Riemerella Anatipestifer Serotypes 2

The outer membrane protein of Riemerella anatipestifer Serotypes 2(RA2) was extracted by ultracentrifugation.The protein bands were separated by SDS-PAGE.The result show that the molecular weight of maior outer membrane protein were 17KD, 32KD, 50KD, 54KD, 74KD,9IKD and 98KD.The 54KD protein showed positive reaction to the aqainst RA2 by westem-blot.the 54KD OMP was purified by electrophoretic elution ,this eluted protein solution was pooled and the protein concentration was detected.Five ducklings were immunized using the purified OMP emulsified with Freund's adjuvant at the age of 8-day old.The second immunization was conducted 14days later.Sem were collected at 7 days interval after the first immunization.The antibody titres were dected by ELISA.The results show that antibody can be tested on the 14th day after vaccination and reach the highest level on the 14th day after revaccination.The mean antibody titre is 640,and the mean tire on the 29th day post revaccination is still 320. These results show that the 54KD OMP extracted from RA2 could induce higher antibody responseRA1 and RA2 inactive bacteria were used to prepare oil adjurant RA1 vaccine.
