Fiber initiaotion is the first phase of cotton fiber development. To clone the genes related to the initiation of cotton fiber is the important foundation for the research of its molecular mechanism.The difference of gene expression between the ovules of Xuzhoul42 (Gossypium hirsutum. L) and those of its fuzzless-lintless mutant on -1DPA and 1DPA were compared by DDRT-PCR. Genes related to cotton fiber development were also cloned by RT-PCR. The results were as follows:1.The CTAB-PVP method was adopted to extract RNA of ovules and high quality RNA was obtained.2. 22 cDNA bands had been recovered by DDRT-PCR. It was proved by Reverse Northern Blotting that seven cDNA fragments were positive. Two of those positive fragments were obtain from Xuzhou142, the others were obtain from its fuzzless-lintless mutant.3. The result of BLASTX had shown that the cDNA clone coded as 03-29-5-1 has 60% identity to nuodulin family protein in Arabidopis Thaliana.4. The cDNA clone coded as 04-05-3-1 isolated from ovule of MXZ142 had 82% identity to Lea4-A gene in Gossypium hirsutum L.5. The 1006bp cDNA fragment was also obtained from ovule of MXZ142. The result of BLASTX had shown that this fragment had 61% identity to hypothetical protein in fava bean. The function of the hypothetical protein in cotton is unknown.6. A 659bp cDNA fragment was cloned by RT-PCT. The result of Blastx had shown that this fragment hasd 63% identity to the members of amino acid transporter family protein in Arabidopis Thaliana. This fragment is deduced as amino acid transporter in cotton.7. Another two clones coded as 11-20-T-1 and 12-23-T-1 had high identity to putative senescence-associated protein (Pisum sativum) and 26S ribosomal RNA gene, respectively.
