In order to investigate the effect of sows immunized with attenuated vaccines via genital tract rout on the local immunity and system immune level, sows were immunized with porcine reproductive and respiratory syndrome (PRRS) attenuated vaccine and Pseudorabies (Pr) attenuated vaccine via genital tract rout, respectively; To investigate the local cell-mediated immunity in genital tract, the number of intraepithelial lymphocyte (IEL ) and mast cells in the uterus and the level of IL-2 in the uterus were detected; To investigate the local humour immunity in genital tract, the level of SIgA in the secreting fluid in the vagina and the number of IgA and IgG secreting cells in the uterus were detected; To investigate the level of the systemic immune response, changes of peripheral blood T cell function and the level of the specific antibody in the serum were detected.1 The effects of different attenuated vaccine of sow on the local cell-mediated immunity in genital tract immunized via genital tractThe number of IEL in the uterus: The result showed the number of IEL in the uterus horn was reduced significantly(P<0.01)after immunization with PRRS attenuated vaccine and the number of IEL in the uterus horn had no significant changes after immunization with Pr attenuated vaccine. The number of IEL in the cervix was reduced significantly(P< 0.05)after immunization with PRRS attenuated vaccine and the number of IEL in the cervix had no significant changes after immunization with Pr attenuated vaccine. The results suggested that the attenuated vaccine may have effect on IELs in uterus.The number of mast cell in the uterus: The result showed that the number of mast cell in the uterus had no significant changes compared to control group after immunization with Pr and PRRS attenuated vaccine. The result suggested that mucosal immune via genital tract didn't affect the number of mast cells in the uterus.The level of IL-2 in the uterus: The results showed that the level of IL-2 in the uterus horn reduced and had no significant changes compared to control group after immunizationwith Pr attenuated vaccine, the level of IL-2 in the uterus horn had no significant changes after immunization with Pr attenuated vaccine; The level of IL-2 in the cervix was reduced significantly(P<0.05)after immunization with PRRS attenuated vaccine compared to control group. The level of IL-2 in the cervix reduced and had no significant changes after immunization with Pr attenuated vaccine. The results suggested that mucosal immune via genital tract can affecte the level of IL-2 in the uterus and the level of IL-2 in the uterus has relationship to the different vaccines.2 The effects of different attenuated vaccine immunized via genital tract on the local humoral immunity in genital tractThe level of SIgA in the secreting fluid in vagina: Double sandwich enzyme-linked was applied to detected the level of SIgA in the secreting fluid in vagina, the result showed level of the SIgA in the genital tract secretion increased significantly(P<0.05)compared to the control group at 3 weeks after immunized by genital tract. The level of SIgA had no significant changes at 4 and 5 weeks compared to that in control group in PRRS attenuated vaccine group. The level of SIgA had no significant changes at 3 4 and 5 weeks in Pr attenuated vaccine group compared to control group. The results suggested that genital tract may be a suitable route for delivering vaccine, but the immune response time in genital tract lasted short. Different vaccines may induce different levels of SIgA in the secreting fluid in vagina.The number of IgA and IgG secreting cells in the uterus: SPA-HRP Immuno-histochemical technique was applied to investigate the distributing and the change of the number of IgA and IgG secreting cells in the uterus. The result showed that IgA and IgG secreting ceils in the uterus were distributed in the lamina propria; the number of IgG secreting cells in the cervix was more than it in the uterus horn, the number of IgA secreting cells in the uterus horn...
