Poplar is one of the important forest trees for timber with fast growth. The research of gene transformation on poplar by gene engineering technique is a effective way for the selection of new species of poplar with insect-resistant, disease-resistant and so on.Several molecule and insect-resistant research were studies on (Populus deltoides X Populus simonii) NL-80106 with land for three years and Populus alba var. pyramidalis with land for one year. PCR analysis of transgenic poplars proved that Bt genes exist steadily in all transgenic plants. But PCR analysis of transgensis on CpTI gene is wrong in size of bands. The leaves from 12 transgenic insect-resistant poplars were analyzed by ELISA and showed that the expressing quantity of Bt insecticidal protein exist distinction .The Bt insecticidal protein of D18, FB51 and FB56 exceed 6700 ng /g fresh leaf, which is 0.090%, 0.036% and 0.034% of plant soluble protein respectively. In the other transgenic clones with single Bt gene, the expressing quantity is 318-4950 ng/g fresh leaf.Insect bioassay indicated that the part of transgenic plants were significantly resistant to larva of Micromelalopha troglodyta , but it did not affect to chrysalides of Micromelalopha troglodyta The study showed that the death rate is 89.2% in 12d to the larva of Micromelalopha troglodyta . Micromelalopha troglodyta were different sensitiveness to Bt poplar of different ages. With worm's age's increase, the sensitiveness is weakened notably. In addition Bt poplar exist the obvious ground time trends to Micromelalopha troglodyta. It is 56.9%~82.4% in June ,which drop to 35.6%~45.1% in July and August and it raise 74.1%-89.2% in September. It exist significantly difference between comparison and transgenic clones to avoirdupois of larva in 8d.Research this experiment by molecule and living beings ,we get the same result , insect-resistant clones have more Bt insecticidal protein. It prove that two kinds of methods can replace each other , learn from other's strong points to offset one's weaknesses.
