Paulownia Witches' Broom caused by phytoplasma is the most serious disease in paulownia. People have no method to control its spread. SHIVA-1 that active site is on membrane is a kind of broadly antibacterial polypeptide. In this study, we confirmed that shiva-1 gene has integrated in the chromosome of the paulownia which still contains phytoplasma offered by Wuhan Institute of Virology by PCR, Southern-blot and dot-blot hybridization. Rooting percent of optimal rooting medium containing l/2MS+0.5mg/IIBA+6g/l active carbon+15g/l sugar is 98%. Humidity and illumination are the main factors that affect the successful transgenic plant. Resistance of transgenic paulownia to witches' broom increased in one year of green house identification: the anaphase average disease grade of transgenic paulownia decreased 1.5 compared with that of forepart; average disease grade of transgenic paulownia is 3 less than that of CK. Symptom of transgenic paulownia witches' broom disappeared slowly; if environment is suitable, transgenic paulownia witches' broom could become serious. The serum titer of BALB/C mouse with SHTVA-1 synthesized manually as antigen is 104. Result of shiva-1 gene expession detected by indirect-ELISA showed that one part of transgenic paulownia has distinct expressed signal, but another part hasn't. Expession difference of foreign gene in transgenic paulownia proved that SHIVA-1 is the main factor of transgenic paulownia that increases resistance to witches' broom at protein level. Some transgenic paulownia blossomed in 2001.
