In this research, high efficient strains were screened for different cultivars of kidney bean grown in vermiculite and soil pots. In order to track the effective rhizobia. Molecular marker were used too.Black kidney bean of Beijing and red kidney bean of Japan planted in vermiculite-pots were inoculated with twenty rhizobial strains respectively. The results showed that four rhizobial strains had significantly positive differences comparing with the controls no inoculated above targets of the aboveground dry weight, the number of effective nodules, the weight of effective nodules, and nitrogenase. The four rhizobial strains are CCBAU03244, CCBAU01367, CCBAU65266 and CCBAU65121. In the soil-pots experiment, there are ten rhizobial strains which include the four effective strains from vermiculite pots, four different cultivars of kidney bean, and four types of soil. The results showed that CCBAU6S121 could significantly increase the four targets in all experiments. CCBAU03244 could significantly increase the four targets in all experiments, except when it planted in the loess. CCBAU01367 and CCBAU65266 could significantly increase the four targets in most of experiments, other strains could only significantly increase some target with some cultivar of kidney bean planted in some soil.In order to track the inoculated rhizobia, RAPD, Box-PCR and reporting gene Lux-AB were used in this research. The results of RAPD and Box-PCR indicated the obvious difference of all the selected rhizobial strains, but die track was fail.The Lux-AB gene has been transferred into CCBAU01367, CCBAU65266, and CCBAU65121 respectively by tri-parental mating. It is pity that illuminant nodules were not detected. So it needs to improve the factors and go to step further detecting it.To detect the stability of effective strain when it was applied to produce, this research found it was stable within the four months by using peat to absorb CCBAU65121, storing in room temperature, detecting the number of colony in interval.
