The test-tuber shoots , its diameters of pseudo-stein were about 2 mm, were employed in this research. The main contents concluded optimization of medium to induce the garlic microbulblet , physiological and biochemical characteristics in microbulblet development and mechanism of dormancy control, which hi order to provide basical theory of microbulblet development and dormancy control .The results were as follows:1.Using Response Surface Methodology to optimize the medium with sucrose .active carbon and salicylic acid , the three fators had the effect on the induction and expansion of microbulblet. It came into conclution that optimizal combination of sucrose,active carbon and salicylic acid,sucrose was 95.8gL-1, salicylic acid was 6.8mgL-1, active carbon was 3.48gL-1, the induction rate of bulblet was 99. 7%, the average weight of it was 402.1mg.2. Using the high concentration of sucrose to induce microbublet with tuber shoots , study the morphological change and the fundament of physiology and biochemistry in bulblet development. During bulblet development, no lateral bud ,only growing point. Its contents of the total sugar kept increasing, contents of the reducing sugar and soluble protein increased earlier, then decreasing. Activities of POD, PPO increased with bulblet expansion then decreased at the end. Changes of CAT activity was conversely, peroxidase isozymes and SDS-PAGE all had more bands, and its dying deeply in the bulblet expansion period.3.Sprouting of graded microbulblet at 25℃ and 15℃ were studied hi the eaxperiment. The results showed :all microbulblet did not sprout at 25℃, the sprouting rate increased with the bulblet weight at 15℃. Sprouting rate of microbulblet with weight greater than 500mg was 100%. To treat microbulblets with low temperature ,GA3, combination of GA3 and low temperarure,flux temperature, the best treatment was combination of GA3 and low temperature, then flux temperature and low temperature, but the single GA3 treatment had no effect on microbulblet sprouting .4. To treat microbulblet with low temperature and combination of GA3 and lowtemperature ,study the fundament of physiology and biochemistry in the bulblet during treatment .The trency of sugar contents , protein contents ,enzyme activities were alike in two treatment, but changes of these indicators were greater in the treatment of combination of GAs and low temperature in contrast with low temperature treatment,especially after ten days. During the treatment , contents of total sugar decreased ; activity of sucrase increased; contents of reducing sugar decreased earlier then imcreased with the days of treatment extended ; contents of soluble protein increased gradually. Activities of POD and PPO increased, but activity of CAT decreased first then increased. The peroxidase isozymes had more bands during after ten days of low temperature treatment , the bands dying deeply in day of 15 and 20 .
