Cytoplasmic male sterility (CMS) is the base of hybrid rice; Meanwhile, it is the good material of studying cytoplasmic genetics, the development of pollens and the interaction between cytoplasma and nuclear, so it is very hot in the academic research recently. Restorer genes in nuclear genome can restores fertility to cytoplasmic male sterile plants, the isolation of restorer gene will help to characterize the molecular mechanism of CMS/Rf system. Here we report the fine map of the nuclear fertility restorer gene for HongLian type cytoplasmic male sterility in rice and construction of its physical map:1. Bulked segregant analysis (BSA) of a BC] population derived from congguang 41A/Milyang 23//congguang 41B was used to map the nuclear fertility restorer gene Rf5 for HongLian (HL) Cytoplasmic male Sterility. The parents and two bulks respectively representing extreme fertile and sterile plants were screened for polymorphism with 22 microsatellite primer pairs onchromosome 10 chosen on the basis of previous research. RM5373 is linked to fertility restorer gene Rf5 at a distance of 0.63cM, and another microsatellite primer: RM1108 was locked at a distance of 0.3lcM to Rf5 in the other direction of chromosome 10; Microsatellite marker RM6737 is co-segregant with Rf5. Closely linked DNA markers will facilitate not only cloning the gene but also marker assistance selection (MAS).2. In an effort to construct the physical map of a fertility restorer gene: Rf5, positive clones were selected from the BAG library IR64 through pool-PCR that the primers were designed according to the known rice Nipponbare sequences between the cloesly locked markers. The physical map is made up of 5 BAC clones. Rf5 gene's physical map forms the base of isolating that gene, and it also will be helpful for the study of the molecular mechanism of fertility restoration.
