Rice has become a model monocot plants because of the accumulation of molecular information for this species, efficiency in transformation ,its small genome ; and the economical importance of this crop.The functional genome research of it is becoming a topic and attractive theme ,based on its completed genomic sequencing.Insertional mutagenesis has the advantage that the inserted element act as a tag for gene identification .And,it is convenience for both forward and reverse genetics research.,So,it play an important role in functional genome research.Insertional mutagenesis using T-DNA,Ac/Ds AND Tos17 are commonly methods to creating a mutant library in rice .We have created a mutant library by means of T-DNA insertion,and analysed the library.The following is the majory results.1 We developed an Agrobacterium-mediated trasformation system in rice cultivar Zhonghua 11,and have a collection of T-DNA tag lines. PCR analysis indicated that tile positive line was about 69.8%. The Southern blot analysis of transgenic plants confirmed that the T-DNA had been integrated into the rice genome and the copy number from 2 to 8 in different line.2We found some types of mutant ,such as dwarf mutants,variation in heading date,mutant of low Tillering ability,mutant of the glume in color and shape .3 We get the T-DNA tag of OSELF3,it is maybe the heading date control gene4We get the T-DNA tag of the AAA+ ATPase family member,it is maybe the dwarf control gene.
