The F1 hybrid between common wheat cv. Shinchunaga x rye cv. Qinling resulted inspontaneous F2 seeds. From the F2 progenies, a new wheat line 99L2 carrying agronomiccharacters from rye and having chromosomes 2n=42, was obtained. The objective of this studyis to detect the rye chromatin of 99L2 by cytogenetics, biochemistry and molecular biologytechniques. The results are as follows: 99L2 had a stable chromosome number with 2n--42. These chromosomes showed normalparing as did in common wheat during meiosis metaphase I of pollen-mother-cell. Combinedwith a consistent agronomic performance and glutenin and gliadin patterns among different99L2 plants, 99L2 was recognized as a stable wheat line in genetics. Glutenin analysis indicated that 99L2 has the 75K gamma-secalins at Sec-2 on ryechromosome arm 2RS. RAPD analysis further proved that 99L2 has genetic materials from 2RSby using specific markers on 2RS. In addition, 99L2 has morphological characters induced by2RS such as tough glume. All these results indicated that 99L2 has the rye 2RS segments. Butno any rye telomeric C-band was detected, thus indicating the missing of rye telomeric C-bandin 2RS. It is surprise that no rye chromosome segments were detected by GISH. It was indicated the possibility that other rye chromosome segments were also introducedinto 99L2. Meanwhile, some wheat DNA sequences in 99L2 were changed in comparison withthat of its donor parents.
