A pair of special PCR primers was designed basing a banana cDNA BR-47 clone which expressed after harvested. Then a 1016bp cDNA segment was gained from the cDNA library of harvested banana fruit by using PCR technology, named BR472. The result of BLASTn showed that there was no homologous sequence of knowed function; the result of BLASTx showed that BR472 had highly homologous with glycogen (starch) synthase gene. The online analysis result of PROPSEARCH showed that BR472 had the highest reliability with PE-PGRS family gene, but the reliability only 32%. The result of conservative sequence analysis indicated that protein encoded by BR472 related with sugar metabolism.The result of Southern blot of banana genome indicated that the gene had one copy in banana genome. The result of Northern blot indicated that the gene was specifically expressed in fruit, and the expression gradually ascended with time going on in harvested banana fruit. It implied the gene was an up-regulate gene in the fruit ripening and intenerating process.
