The scerotinia sclerotiorum, a main disease of Brassica napus especially in YangZi drainage area, has been attached importance on its mechanism by many scientists since last century. Nevertheless, we have not got much inspired improvements because of the limited methods of investigation and the extraordinary complicated nosogenesis. Whereas the difficulties they met, people's work has stayed in the situation of biochemical observation and preliminary genetic study, which have made a chokepoint of resistant plant breeding.The first proposition of this paper is to display, by using CZE ( Capillary Zone Electrophoresis) , the differences of proteins extracted from leaves pre-treated with scerotinia sclerotiorum and its controls. We have successfully detected the differences in our experiment materials with this method. The optimal process have demonstrated that the results are pretty good under pH2.6,25-30C,25-30kV, and the precision of this determination is also encouraged, with successive six runs reproducibility of one assigned peak being typically less than 1%R.S.D (Relative Standard Deviation).At the meantime, we have displayed the differences of ZhongYou821 and its backcross with 888(BC5 ZhongYou821 x888) on their transcriptional level of RNA by 72pairs primers(26 random primersx3 anchored primers)using DDRT-PCR, unfortunately we have not got any apparent differences.In this study, we also designed three pair of primers according to some aligned conserved homologous sequences of resistant genes and their cDNAs (PAL, SOD-Cu/Zn, SOD-Fe) which have been cloned from various plants. The result shows that every pair of primer can amplify one main band from ZhongYou821 genome or cDNA under a suitable condition. After recovering, cloning and sequencing those bands, finally, the analysis of BLASTN in Genebank reveals that the bands possibly have a relation to resistance of plant diseases.
