| Agronomic traits' and cytological characteristics' research were carried out on thematerials of Leymus mollis, Tritileymus, a variety of common wheat, Yannong15 and aTriticum-Leymus translocation line, Shannong 0096 in this study preliminarily. On thebase of that, the Triticum-Leymus translocation line, Shannong 0096 was study further byusing the technology of genome in situ hybridization (GISH) and random amplifiedpolymorphic DNA (RAPD) and the gene resistant to yellow rust from Leymus molliswhich carried by Shannong 0096 was identified and analyzed by molecular marker. Theresults were as follows. 1.The analysis results of the chromosome configuration of these materials showedthat the chromosome number of Leymus mollis' root tip cell was 2n=28, the chromosomenumber of Octoploid Tritileymus' root tip cell was 2n=56, and that of Shannong 0096 andYongnong 15's was 2n=42, respectively. And the chromosome configuration at PMC MIof Leymus mollis was 2n=14â…¡, the chromosome configuration at PMC MI of OctoploidTritileymus was 2n=8â…¡, and that of Shannong 0096 and Yongnong 15 was 2n=21â…¡,which revealed that the germplasm was stable in cytology. 2.Mean chromosome configuration of hybrids F1 between Shannong 0096 andwheat parent at PMC MI was 2n=19II+1IV,which primarily indicated that shannong0096 was a wheat-Tritileymus translocation line. The results of GISH with the probe ofLeymus mollis genome DNA, further proved that Shannong 0096 was a wheat-Leymusmollis translocation line with a small Leymus mollis fragment which reciprocallytranslated in the short arms of a pair of wheat chromosomes. 3. 300 random primers were screened among Shannong 0096 and its parents byusing RAPD technology. And 8 primers of them can produce polymorphic DNA bandsbetween the materials, which proved that Shannong 0096 confered genetic ingredient ofLeymus mollis. 2山东农业大å¦ç¡•士å¦ä½è®ºæ–‡ 4. Artificial inoculation of yellow rust was performed to Shannong 0096 and itsparents and the results of resistance investigate revealed that Shannong 0096, Leymusmollis and Octoploid Tritileymus were immune to yellow rust. RAPD analysis to the F2mapping population, obtained from the cross between Shannong 0096 and Chinese Spring(C.S.), showed a RAPD marker linked to the gene with resistance to yellow rust wasselected and designed as OPTA20485. |
