| In this study, the whole process of propagation of floral organ of lily (L. Asiatic Hybrid) in vitro was conducted. The main results are showed as follows:There had differences in callus induction on the various media. Callus of receptacle was induced on MS media, but not on other media. Among floral receptacle, petal, Style and peduncle, 6-BA and NAA had better effect on induction of callus than other auxins and cytokinins. These four explants could all be induced to produce callus, of which the induction rate of floral receptacle was the highest. Floral receptacle had the highest induction rate of callus on the media of MS+6-BA1.0 mg-L-1+NAAO-lmg-L-1. Adventitious bud were induced directly from floral receptacle on the media of MS+6-BA1.2mg-L-1 + NAAO.l mg-L"1. Adventitious bud obtained from these calli quite well. The media of MS+6-BA1.0+NAA0.1 had the best effect on proliferation of adventitious bud. The media of 1/2MS+NAA0.2 had the better effect on rooting induction. Transplanting rooted shoots to substrate composed with l/3perlite, 1/3 vermiculite and 1/3 humous soil, survival rate of shoots was increased to 98%.Soaking explants in the solution of 50 mg-L-1Vc before inoculation and adding 150 mg-L-1PVP to medium during subculture could control browning very well.Expansin crude extraction of soybean (Glycine max) had a higher acceleration to seedings growth in vitro of lily, and the optimal concentration of expansin was 3.0g/L.In various stage of induction, there was obvious influence on development and growth of lily seeding by reducing the content of sugar gradually. With the low sugar content and the complementarity of CC>2, there was a remarkable effection on increase of the autotrophic ability of lily. |
