Pholiota adiposa (Fr.) Quel . is a kind of edible and medicinal fungi.It belongs to Strophariaceae, Agaricales, Homobasidiae, Basidiomycetes, Eumycota.27 and 104 monokaryons were isolated from two sporocarps of Ph adiposa by spore-splashing and dilution isolation. By using three-cycle mating system ,the crossing character of Ph. adiposa was studied and the result suggested that Ph. adiposa is a kind of basidiomycete with quadrupole heterothallism. The study also showed the proportion of the different mating type is patency deflection.RNA of different period including mycelium , anlage, bud and fruit body (stem of mushroom and lid of mushroom) was extracted and treated by RT-PCR using anchored oligo-dT primers and arbitrary decamers, and the DNA fragments were differential displayed in agarose gel electrophoresis. There are 32 differential display in 78 pictures of electrophoresis. Six intreseted fragments were isolated , reamplified .purified , cloned and mensurating sequence .Five were succeed . After blasting in GeneBank we identified T,,G B0322 cDNA fragment is similar to oxysterol-binding protein.This show that may be a gene fragment associated with differentiating developing of Ph adipose.
