Enzyme-linked immunosorbent assay for TGE yolk antibody was established successfully. TGE yolk antibody and high immune powder by praying dry were prepared by immuning yi-sha hens with two TGE-PED vaccines of death and attenuating. Antibody value in serum and egg yolk was detected by indirect ELISA in order to study dynamic change of ebbing and flowing of antibody in serum and egg yolk. Antibody value of TGE yolk powder was detected, too ;At last, changes of thyroid hormone and IL-2 in immuned hens serum were discussed.1. The development of indirect ELISA for TGE yolk antibodyThere were some reports about detecting TGE antibody in pigs by ELISA, but none in TGE yolk antibody. In the development of indirect ELISA for TGE poly-cloned yolk antibody, the antigen was TGE virus particle and its coating concentration was determined as 82ng/ml by chess-board experiment, then the stability of ELISA was tested. The results demonstrated that when antigen coating concentration was 82ng/ml, its reaction to positive serum was well special, little background for negative serum(value below 1:20); Stability test indicated the pretty stability of ELISA: for example, inter-plate variation(n=3) were 2.9%(positive control), 10.8%(negative control); inter-well variation were 5.3%(positive control), 3.2%(negative control).2. The variation of antibody in serum and yolk of hens inoculated with two kinds of vaccines one after the other and the preparation of TGE yolk antibody.Eighteen healthy hens of 200 days old were divided into control and experiment groups according to weight, 9 hens each group and one hen in one cage, all were in the same feeding condition. No TGE antibodies in experiment hens serums were detected positive by AGP after third immune with TGE -PED dead vaccine of pigs; Then changing to TGE-PED dead vaccine of pigs, five in the experimental hens serums were detectedpositive by AGP; Serum specimen of control and experiment groups were collected at separate times(0, 30, 83, 133, 155, 171, 185, 208, 232 days) and yolk specimens were collected at separate times(171, 179, 185, 195, 199, 208, 232, 266, 295, 326, 361 days), comparing dynamic change of ebbing and flowing of antibodies in serum and egg yolk. The results demonstrated that TGE-PED attenuated vaccine had better immunogenicity than TGE-PED dead vaccine; Antibody concentrations in the serums, was a little higher or equal and earlier reached pinnacle comparing those in the yolks; Antibody titer in the yolk could maintain two months at the pinnacle(over 14) and only 10 days at the peak(over 16); Antibody titer in the yolk did not increase after sixth immune of the attenuated vaccine;Besides, two batches of TGE yolk powders of praying dry had titers of over 1:4000 and 1:8000 separately.3. The effect of immuning hens with two kinds of vaccines one after the other on thyroid hormone and interleukin- II in the bloodT3, T4 and IL-2 concentrations in the serums of control group and experiment group were mensurated by radiative-immune assay at four different times: 0 day(before immune), 83 days(after the third immune of dead vaccine), 133days(after the first immune of attenuated vaccine) and 232 days(after the fifth immune of attenuated vaccine). The results demonstrated that T3 and T4 concentrations of serum was significantly different between control group and experiment group(p>0.05), serum IL-2 concentration of experiment group significantly increased comparing to control group and exhibited the trend of edding.
