Alfalfa (Medicago sativa L.) is one of the most important legume forages in China . In this experiment, Medicago sativa L. cv. 'Zhongmu No.l' was used as the material for studying cold resistance, salt tolerance, and leaf protein extraction. The results are as follows:1. Germination percent of cv. 'Zhongmu No.l' decreased with the increasing of salt concentration., and the percent changed greatly within the range of salt concentration at 0.6%~0.8%. Seedling growth of cv. 'Zhongmu No.l' can be promoted when the salt concentration is low. The concentration of 0.6% NaCl solution could be taken as the appraisal concentration of salt tolerance for seeds of cv. 'Zhongmu No.l', and seedling height and rdicle length can be taken as the indexes of salt tolerance for cv. 'Zhongmu No.l' during germination. .2. Seedling growth in height was restrained obviously and the symptom of salt injuring was more serious with the increasing of salt concentration and the prolongation of disposal time. Seedling height can be used as the index of appraising salt tolerance for cv. 'Zhongmu No. 1' dyring seedling period. Root length, however, could not be taken as the indexes of salt tolerance for cv. 'Zhongmu No.l'. Fresh weight and dry weights can be also as the index of salt tolerance for cv. 'Zhongmu No.l'. The changing law of cell membrane permeability was the same with salt concentration, so it can be as the index of salt tolerance for cv. 'Zhongmu No.l'.3. Spraying exterior ABA under the salt stress could produce some effects on cv. 'Zhongmu No.l'. There was no distinct affection of ABA on seedling height, dry weight and cell membrane permeability when salt stress unexisted, however, it produced different effects under various salt concentration, such as decreasing of salt restriction on growing, increasing of dry weight, reducing of cell membrane permeability and increasing the salt tolerance of cv. 'Zhongmu No.l'. This is basically identical with the results of some former researchers on wheat, barley and maize.4.The result showed:Under low temperature, at 12th hour the activation of SOD POD CAT of cv. 'Zhongmu No.l'leaves was the highest, and at this time cv. 'Zhongmu No.l'had the highest cold resistance characteristics;Afterwards ferments activation descented swiftly,the cold resistance ability of cv. 'Zhongmu No. 1 'droped too.5. Leaf protein extration shows that the way of fermentation produced the highest extracting percent, microwave way was a bit low perhaps for the reason of its too high power.
