A new model containing artificial and natural cellulose, hemi-cellulose, has been developed for enumeration of straw-utilizing bacteria in soil. The model not only could rapidly and effectively isolated cellulose-utilizing bacteria from a lot of samples, but be thought over microbial ecology in situ. By the model, J-1211, J-1212 were isolated from rotted rice straw of Agricultural Science Academy of JiangSu provincial, and HA-2, HC-6 were isolated from rotted fallen leaves of ZiJin Mountain,respectively.According to bacterial configuration, physiology and biochemical characters, J-1211, J-1212, HA-2 and HC-6 were identified as Paenibacillus spp. And profile of denaturing gradient gel electrophoresis(DGGE) showed they would belonged to different special.The isolated strains could grow on agar of pH 9 and salt content 5%, and could tolerant some heave metal ions, such as Cd2+, Cu2+, Ni2+and Zn2+, pesticides and herbicides, which would be helpful to the strains living in soil. J-121 1, J-1212, HA-2 and HC-6 were harmless for rice, chili and cotton. The isolated strains could prevent Xanthomonas oryzae, Fusarium graminearum, Fusatium moniliforme from growing by nutrition competition.The P-l,4-glucanases of HC-6 and J-1211 have capacity of resisting alkali. The endo-P-l,4-glucanases activities of HC-6 and J-1211 could maintain 60.2% and 43.2% of max enzyme activity at pH 11.40. The endo-P-1,4-glucanases of J-1212 could maintain 89.2% of max enzyme activity at 50C. The amylase activities of HC-6. J-1212. J-121 land HA-2 were 21.6Uml-1, 18.6Uml-1, 13.lUml-1 and 7.11Uml-1, respectively. The pectase activities of J-1211. J-1212, HA-2 and HC-6 were 23 .05umolmin-1ml-1. 21.48Uml-1, 21.13Uml-1 and 21.05Uml-1, respectively.Through single-factor and multiple-factors experiment, the optimum fermentation medium of J-1211 was confirmed as starch 0.8 % . sugar 0.2 % . NaNO3 1.5 % , yeast extract 2.5 %, NaCl 0.1 % . KH2PO4 0.1 % . CaCl2 0.01 %. MgSO4 0.03 %, FeCl3 0.001 %, H2O 1000ml, 28, pH8.5; the optimum fermentation medium of J-1212 was: starch 0.8 %, sugar 0.2 %, NaNO30.5%, yeast extract 2.5%. NaCl 0.1 %, KH2PO40.1%, CaCl20.01 %, MgSO4 0.03 %, FeCl3 0.001 %, H2O 1000ml, 28,pH8.0; the optimum fermentation medium of HA-2 was: paste 0.5% , glucose 0.5% , NaNO21.5%, (NH4)2SO2 1.5% , NaCl0.01%, K2HPO40.1%, CaCl20.01%, MgSO40.03%, FeCl3 0.001%, H2O 1000ml, 37,pH7.5; the optimum fermentation medium of HC-6 was: paste 0.5% , glucose 0.8 %, NaNO21.5%, (NH4)2SO2 1.5% , NaC10.1%, K2HPO40.1%, CaCl20.01 %, MgSO40.03%, FeCl30.001%, H2O 1000ml, 37,pH7.5.The combination of J-121K J-1212 , HA-2 and HC-6 could decompose 88.3% of rice straw in 5 days. In Cd2+ contamination soil, the result of cellulose-decomposed of JM combination was better than that of other bacterial agents and tow controls. The ratio of cellulose-decomposed of JM combination was 87.57% at the end of culture period. The color of rice straw turned into brown and black and plant tissue was decomposed obviously.In Taihu paddy soil of water holding content 40%~60%, the cultural temperature was the most important factor for rice straw decomposed, and the degree of rice straw decomposed at 20~28C was better than that at -2-10 C. But under irrigation basin, temperature might not important factor. JM combination could prove the content of available K and available P were increasing in Taihu paddy soil and Jiangxi red soil .The changes of available K and available P content in soils could have close relations with the condition of rice straw returned filed. In soils of water holding content 40%~60%, available K and available P in paddy soil would greatly increase 55.1% and 9.27 times, available K and available P in red soil would greatly increase 1.27 times and 92.6%, respectively. While under irrigation basin, available K and available P in paddy soil would greatly increase 84.9% and 2.77 times, available K and available P in red soil would greatly increase 58.8% and 31.2%, respectively.JM combination could improve invertase activity and cellulase activity in paddy soil and red soil.
