The Elytrigia Desv. is rennial grass of Graminaea. It is a good specie of grass for setting up artificial grassland and improving natural grassland. This study selected and authenticated 20 different Elytrigia repens L. and E. intermedia Nevski germplasm, which is from home and abroad, in the glasshouse and field drought, selected out primers with molecular markers E. repens L. and E. intermedia Nevski by using SSR, in order to provide science theories basis and base to further study E. repens L. drought resistance gene molecular marker, discover excellent drought resistance gene source, fast and efficiently select with the molecular marker of drought resistance gene catena for E. repens L. and E. intermedia Nevski. The pot experiment was conducted to study E. repens L. and E.intermedia Nevski at the seedling period in glass house under drought stressful circumstances, this experiment measured four drought resistance indexes: relative water content of the leaves, relative electrical conductivity rate, free proline accumulation and content of malonyldialdehyde, used cluster-analysis system to classified cluster to date, could be classified into 3 drought resistance ranks: the strong drought resistance, the medium drought resistance and weak drought resistance. Field experiment used random blocks experiment design, on the condition of natural drought, we measured the morphological indexes: heights leaf width and physiological indexes: relative water content of the leaves, relative electrica conductivity rate, free proline accumulation and content of MDA that were related to the drought resistance in different growing stage, synthesizing to assess the drought resistance according to the average of subordinate function. According to the experiment results of seeding stage, which used SSR reaction system and PCR reaction hot process, as template that E. repens L. and E. intermedia Nevski DNA has different drought resistant ranks, above SSR reaction system was used in selecting 240 lines primer of twelve groups after five turns, for eight pieces of E. intermedia Nevski. We selected out primer wm264 that have produced stable polymorphic DNA fragments between 242bp and 309bp; for twelve pieces of E. repens L., we select out primer wm314 that have produced stable polymorphism DNA fragments between 217bp and 242bp, so we believe that the two primers can be looked as special marker of different E. repens L. E. intermedia Nevski drought resistance markers.
