The abortifacient effect of different dosages of bromocritine on the mouse and the role of monomer ingredients of scutellaria baicalensis Georgi: Baicalin, Baicalein and Wogonin are investigated in this study. Via in situ hybridization, western blotting and measurement of the contents of IFN-γ , progesterone in serum and uterus lysate, the possible anti-abortive mechanism of Monomer Ingredients of scutellaria baicalensis Georgi is explored in order to explicit the maternal-fetal immunity and to look for safer and Iess-residued drug for early embryo loss. The pregnant mice are divided into 7 groups: i.e., control group, ethanol group and 0.025mg, 0.05mg, 0.075mg, 0.1mg, 0.2mg of bromocritine groups. On day 5~7 of pregnancy subcutaneous injection is carried out respectively, pregnancy results are observed on day 10. The results show that the abortion rate of 0.075mg group is 75%, being in accordance with the miscarriage model. The mice are then given orally distilled water, scutellaria decoction, Baicalin, Baicalein, Wogonin respectively on day 1~7 of gestation. All the mice are injected subcutaneously with bromocritine at the dose of 0.075mg per mouse on day 5—7 of pregnancy, and killed on day 10 of gestation. The miscarriage rate of Baicalin is the lowest among all the treated groups, indicating the best anti-abortive effect. Then the mice are administrated orally on day 1~7 of gestation with different dosage of Baicalin(0.1mg, 0.5mg, 1mg), injected subcutaneously with 0.075mg bromocritine on day 5—7, sacrificed on day 3 , 5 , 7 , 10, 12 by removing their eyes. The serum and the right uteri lysate are collected. The contents of Th1 cytokine IFN- Y are assayed through ELISA. The expression of IFN- Y is detected through western blotting. The left uteri is fixed by Bouin's solution for in situ hybridization to detect the expression of IFN- γ . Western blotting results show a clear band at 45kD. in situ hybridization results reveal more granules in the membrane propria. The granules are significantly greater in 0.075mg and 0.1mg bromocritine groups compared with Baicalin treated mice.
