Cotton was one of the most difficult regenerative plants. Compared to other crops, cotton tissue culture was lagging behind. Over these years, a great deal of progress was made in the course of cotton tissue culture and the upland cotton regenerated system had been built in some labs. But these studies were mainly concentrated on a few varieties, particularly some model varieties. And there were still some problems to be solved. For example, the rate of germination on embryoids was low, the abnormal embryos proportion was high and the cycle time of regeneration was long, and so on. Therefore, there was a need to broaden the number of regenerable cotton varieties and found new cotton plant regeneration system.In order to broaden the genotypes range of cotton tissue culture and increase the number of regenerative plant. The objective of this work was to study the somatic embryogenesis, plant regeneration system and suspension culture in Gossypium hirsutum zhong521 which was the variety that resisted blast and cyanosis, had better integrate characters, had big cultivating areas and was not studied by some researcher before, the results are as follows:1 Gossypium hirsutum zhong521 was studied on somatic embryogenesis. Further, the plant regeneration system of solid and liquid tissue culture was established on the basis of the former studies and the cycle of regenerative plant was shortened 2 months.2 Effects of several phytohormones and their combination were studied on callus induction. The result showed that the effect of IBA on callus induction was better than IAA, 2,4-D was the necessary hormone for callus induction and the induction of embryogenic callus needed low concentration of 2,4-D, callus were inclined to indurate by high concentration of KT.3 The impact of glutamine and asparagines in solid culture and liquid culture were studied. The result showed that 1.0g/L glutamine and 0.5g/L asparagines had acceleration for embryogenic callus inclination and 0.5 g/L glutamine made for embryoids information and 2 g/L glutamine was benefit to plant regeneration.4 Study the problem of brown in the course of liquid culturing, and find that the method of solving brown that did not been reported in the past, that is to say, it can...
